ANTIOXIDANT ACTIVITIES OF ALLIUM PHRYGIUM AND ALLIUM OLYMPICUM

This study was aimed to evaluate the antioxidant potential of water and ethanol extracts from aerial and bulb parts of two endemic Allium species, A. phrygium Boiss. and A. olympicum Boiss. Antioxidant capacities of the extracts were determined by measuring the total phenolic (as gallic acid equivalents, GAE) and flavonoid compounds (as catechin equivalents), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effect and ferric-reducing antioxidant power (FRAP). It was found that the DPPH radical scavenging activities of the extracts increased in a dose-dependent manner and the activities correlated with total phenolic contents. The highest total phenolic contents and the strongest DPPH radical scavenging effects (% inhibition at 0.8 mg/mL concentration) were obtained with the
ethanolic A. phrygium extracts from aerial part (16.59 ± 0.72 mg GAE/g extract and 86.98 ± 3.25%, respectively) and A. olympicum water extract from aerial part (14.22 ± 1.39 mg GAE/g extract and 84.02 ± 4.37%, respectively). The FRAP values (|M of Fe2+ equivalent) of the extracts at 0.3 mg/mL concentration varied from 89.67 ± 3.83 |M (ethanol extract of A. phrygium, aerial part) to 54.98 ±A0.80 |M (water extract of A. phrygium, bulb part). The results showed that the ethanolic A. phrygium extract from aerial part has the best antioxidant capacity. However, water extracts from bulb parts of both A. phrygium and A. olympicum showed no detectable antioxidant activity in this study. Data obtained from the experiments suggest that these Allium species could be a potential source of natural antioxidant.
IN VITRO ANTIOXIDANT, DNA PROTECTION CAPACITIES AND CYTOTOXIC ACTIVITIES OF ALLIUM KURTZIANUM EXTRACTS

Allium kurtzianum [Ascherson & Sint. ex] Kollmann, known as wild garlic in Turkey, is a herbaceous herb of the Amaryllidaceae family. A lot of plants of Allium genus are consumed as food sources and it is used as folk medicine since ancient times. A. kurtzianum is an endemic plant found on slopes of Kazdagi in Turkey. The aim of this study was to investigate the antioxidant, DNA protection and anticancer activities of A. kurtzianum aerial and bulb parts, comparatively. The antioxidant activities of A. kurtzianum methanol extracts were identified by using 2,2'-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, ferric-reducing antioxidant power (FRAP) assays along with the determination of total phenolic and flavonoid compounds in the extracts. DNA damage protection capacity was determined using DNA Nicking assay. Total phenolic and flavonoid compounds were calculated using the Folin-Ciocalteu and the aluminum chloride methods, respectively. In vitro cytotoxic activity of the extracts was identified using 3-(4,5-dimethylthiazol-
2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) reagent on various cancer cell lines. The aerial and bulb parts of A. kurtzianum showed DPPH scavenging acitivity, with EC50 values of 4.63 + 0,13 mg/mL, 12.42 + 0.087 mg/mL, respectively. The reducing activity of the aerial parts (3.39 + 0.090 mM Fe+2) was higher than that of the bulb parts (1.77 + 0.067 mM Fe+2). The antioxidant activities of the extracts can be attributed to a high content of phenolic and flavanoid compounds. The extracts also dose-dependently protected pBR322 plasmid DNA from hydroxyl radicals. The aerial parts of A. kurtzianum exhibited cytotoxic effect against prostate (IC50 0.096 mg/mL), lung (IC50, 0.059 mg/mL), endometrial (IC50, 0.169 mg/mL) cancer cell lines. On the contrary, the bulb parts did not. In summary, a methanolic extract from A. kurtzianum showed potent antioxidant/DNA protecting effects and anticancer activity against some cancer cell lines.
