CHOLINESTERASE INHIBITORYAND ANTI-OXIDANT ACTIVITIES OF THREE SPECIES OF DRACOCEPHALUM

Dracocephalum species are mentioned in Iranian Traditional Medicine for enhancement of cognitive performance.Recently, the Dracocephalum species and their chemical constitution have attracted much attention due to their various biological effects [1].

This study analyzed the AChE and BChE inhibitory activities as well as anti-oxidant effects of five different solvents extracts of three Iranian Dracocephalum species (D. kotschyi Boiss., D. multiccauleae Montbr. &Auch., D. polychaetum Bornm.)

The extractions were performed stepwise usinghexane, chloroform, ethylacetate (EtOAc), methanol (MeOH) and water. AChE and BChE inhibitory assays were measured by a microplate assay [2]. Anti-oxidant activities of the extracts were evaluated using total phenol, DPPH, and FRAP assays [3, 4].

The result showed the most active anti-oxidant activity for the EtOAc extract of D. multicauleae and the MeOH extract of D. kotschyicorrelating withthe highest amount of phenolic compounds among the tested extracts.

The IC50 values for DPPH assay of EtOAc extract of D. multicauleaeand MeOH extract of D. kotschyi were determined as 25 and 23 ig/ml, respectively. The FRAP assay showed the activity for the mentioned extractcs as 230 and 80 mg FSE/g extract, respectively.

The strongest AChE inhibitory activity (79.0% at 100 ig/ml) was measured for the EtOAc extract of D. multicauleae, while the same solvent extract of D. Polychaetum showed the most potentBChE inhibition (62.7% at 100 ig/ml).

It can be concluded that Dracocephalum species are potential natural candidates for the improvement of cognitive disorders.

Rosa damascena Mill. (Rosaceae) exhibited several pharmacological properties such as anti-HIV, antibacterial, antioxidant, antitussive, hypnotic, antidiabetic, and relaxant effect on tracheal chains [1]. It is cultivated in Turkey, Bulgaria, France and India due to its essential oil. After the methanol extract of the petals was prepared, n-butanol, methylene chloride and water fractions were obtained from methanolic extract by partitioning. In this study the sun protection potentials, antioxidant capacity and radical scavenging activity of the extracts were examined spectrophotometrically. SPF values were calculated using Mansur equation. According to the results n-butanol fraction showed the potent sun protective property (16.74) at 200 ig/ml.

Radical scavenging activity of extracts was determined according to the DPPH and ABTS assays. n-butanolic (IC50= 43,79|ig/ml; IC50= 57.53 ig/ml) and methanolic

(IC50= 63.81 ig/ml; IC50= 79.87 ig/ml) extracts showed the best radical scavenging activity in both assays, respectively.

Antioxidant capacity tests were performed by CUPRAC, FRAP and Phosphomolibdate assays. As a result of the CUPRAC, FRAP and Phosphomolibdate experiments, it was determined that the n-butanol fraction showed the best antioxidant capacity.

According to the results n-butanolic extract exhibited potent sun protection effect, antioxidant capacity and radical scavenging activity. From this viewpoint n-butanolic extract of R. damascena petals can be used for antiaging purposes.
