Study on chemical constituents from roots of Sambueus williamsii Hance

Abstract: To study the chemical constituents from roots of Sambucus williamsii Hance. The chemical constituents were isolated and purified by repeated silica gel column chromatography, ODS column chromatography, preparative HPLC. Their structures were identified on the basis of ESI-MS and NMR spectra. Eleven constituents are isolated from 50% ethanol extract of Sambucus williamsii Hance, They are identified as7a-O-ethylmorroniside (1), 7p-O-ethylmorroniside (2), 7-dehydrologanin or 7-ketologanin (3), 7-formyloysecologanin (4), ligstroside (5),(+)pinoresinol-8-O-P-D-glucopyranoside(6), (7S,8R)-4,9,9'-trihydroxy-3,3'-dimethoxy-7,8dihydrobenzofuran-1'-propanolneolignan(7), (7S,8R)-4,9-dihydroxy-3,3'-dimethoxy-7,8dihydrobenzofuran-1'-
propylneolignan-9'-O-P-D-glucopyranoside(8),4,7,9,9'-tetrahydroxy-3-methoxy-8-O-4' isolignan-3'-
0-P-D-glucopyranoside (thero) (9), arctiin(10)and 3-methoxy-4-(2-glycerol)- phenylpropanol(11).

Sambucus williamsii Hance belong to the family Caprifoliaceae, It is also called "Jie-gu-mu", "Tie-gu-san", "Shu-jin-shu"[1]. S. williamsii Hance is a deciduous shrub or small tree,widely distributed in China.The root bark of this plant is a folk medicine with a long history of use in China for the treatment of fractures and osteoporosis. Studies showed that triterpenoids, flavonoids, lignans and the iridoid glycoside are major constituents of S. williamsii Hance [2].In our present work, investigation of its ethanol extract led to get 11 compounds isolated. In this paper, we present the isolation and structural characterization of 11 compounds on the basis of the interpretation of spectral data, including 1D, 2D NMR and HRESIMS data.
Materials and methods
1. General: In the following methods of investigation, we use Finnigan LCQ LC-MS, Bruker-400, 2695-2996-HPLC, Delta-600-2487-HPLC, silica gel column, ODS column, Silica ge160F254, Rp-18, Hypersil ODS H (4.6x200mm,5^m),Senshu Pak PEGASIL ODS H (10x250mm,10^m), Hypersil ODS II (20x300mm,10^m).
2. Plant Material: The root barks of S. williamsii Hance were collected in August 2008 from the Fangzheng district, Heilongjiang Province, China, and identified by the author Zhen-Yue Wang. A voucher specimen (20080813) has been deposited at Heilongjiang University of Chinese Medicine, Harbin, China.
3. Extraction and isolation: The dried root barks (5.0 kg) were extracted with 95% EtOH (2 x 10 L) for 2 h. The EtOH extracts was concentrated under reduced pressure and fractioned on an AB-8 macroporous resin column(8 x 60 cm) with H2O, 50% and 95% EtOH-H2O to get three fractions. The 50% EtOH-H2O fraction (103.0 g) was isolated by repeated silica gel with a gradient of CH2Cl2/MeOH (15:1—>1:1) solvents as eluents to afford I fractions. Fraction IH (20 g) and Fraction ^(30g) continue to be isolated and purified by silica gel chromatography , ODS column chromatography and preparative HPLC. Finally, we obtain compound 1(9mg),compound 2 (11mg),compound 3 (8mg),compound 4 (11mg), compound 6 (9mg),compound 8 (12mg), compound 9 (10mg),Fraction H(20g)is isolated and purified by silica gel chromatography, We obtain compound 5 (7mg), compound 7 (12mg),compound 10 (30mg),compound 11 (9mg).
Results and discussion

0-P-D-glucopyranoside(6)[8],(7S,8R)-4,9,9'-trihydroxy-3,3'-dimethoxy-7,8dihydrobenzofuran-1'-propanolneolignan(7)[9],(7S,8R)-4,9-dihydroxy-3,3'-dimethoxy-7,8dihydrobenzofuran-1'-propylneolignan-9'-O-P-D-glucopyranoside(8)[9],4,7,9,9'-tetrahydroxy-3-methoxy-8-0-4'isolignan-3'-0-P-D-glucopyranoside(theroX9)[10],arctiin(10) [11],and3-methoxy-4-(2-glycerol)-phenylpropanol(11)[12].
