HYPOGLYCAEMIC ACTIVITY OF TWO CUCURBITACINS, ISOLATED FROM IBERVILLEA LINDHEIMERI, AND THEIR ACETYLATED DERIVATIVES

Ibervillea lindheimeri (A. Gray) Greene is a perennial climbing vine from the Cucurbitaceae family [1]. It is one of the 6 species of genus Ibervillea which includes I. sonorae; used in traditional medicine for the treatment of diabetes mellitus. Although hypoglycaemic properties of I. sonorae are well documented no other studies have been made to assess the effect of other Ibervillea species over glycaemia. A chemical study of ethyl acetate extract (EtOAc) from I. lindheimeri's root showed the presence of cucurbitacin-type triterpenes [2]. Cucurbitacines are known for its biological activities such as hypoglycaemic activity [3, 4]. In this study, we evaluated the hypoglycaemic effect in vivo of the EtOAc extract of I. lindheimeri'sroot as well as its most important principles 23,24-dihydrocucurbitacin D (1) and 2-O-p glucopyranosyl 23,24-dihydrocucurbitacin D (2). The acetyl derivatives 2,16-diacetyl 23, 24-dihydrocucurbitacin D (3) and 2-O-p(2', 3', 4',6' tetra acetyl) glucopyranosyl-16 acetyl-23,24-dihydrocucurbitacin D (4) also were tested. Our

results showed that the extract, the aglycone (1) (83 mg/kg), the glycoside (2) (500 mg/kg) and the acetyl derívate (3) reduce the glycemia in CD1 diabetic mice compared to diabetic control (P < 0.001, 0.05 or 0.001 respectively vs. diabetic mice + vehicle). In conclusion we showed that the isolated cucurbitacins are responsible for the hypoglycaemic properties of I. lindheimeri extract. These results indicated I. lindheimeri as a new plant with antidiabetic properties as well as source of antidiabetic compounds.

Many plant species are used extensively for the treatment of various diseases, including atherosclerosis, cardiovasular diseases, inflammation, colds, bronchitis. The bioactive phytochemical constituents present in the plants play an important role in improving the antioxidant defenses of the organism against free radicals and the prevention of pathological conditions.

The purpose of the present study is to determine the antioxidant activity of 5 plant species: Coriandrum sativum L. (CSF), Illicium verum Hook. f. (IVF), Mentha spicata AL. (MSH), Petroselinum crispum (Mill.) Fuss (PCH) and Zingiber officinale Roscoe (ZOR).

The antioxidant activity of the ethanol extracts was evaluated with the 2,2'-diphenyl-1-pycrylhydrazyl (DPPH), ferric reducing antioxidant power (FRAP), non-site-specific- (NSSOH) and site-specific hydroxyl radical-2-deoxy-d-ribose degradation (SSOH) assays. Additionally, total phenolic content was determined by Folin-Ciocalteu method (TPC), and total flavonoid

content (TFC).

All plant extracts showed significant antioxidant activity, with Zingiber officinale exhibiting highest antioxidant potential (IC50 0.069 mg/mL). The FRAP capacity for the tested extracts is decreasing in the following order: ZOR > IVF > MSH > CSF > PCH. Inhibition ofthe production of hydroxyl radicals tested with the NSSOH and SSOH assays determined that ZOR and IVF manifested highest potential of hydroxyl scavenging activity. TPC and TFC values ranged from 50 - 415.8 mg gallic acid equivallent/g crude extract and from 10.6 - 54 mg quercetin equivallent/g crude extract, respectively. ZOR, IVF and MSH showed very high total phenolic content that corresponds to their antioxidant potential, but further investigations on more samples are needed to determine the correlation.

Ethanol extracts of Zingiber officinale Roscoe and Illicium verum Hook. f. could be considered as potential sources of bioactive compounds with strong antioxidant activity. The data obtained with the in vitro antioxidant assays suggest a high potency, but further testing is needed to determine the bioactive compounds responsible for the antioxidant potential.
