Objective To reach the effect of Sambucus glycosides tablet on bone fracture healing following bone injury and its mechanism. Methods Fractures were made in the left tibiae of male New Zealand White rabbits. The fractures were made using the technique of incomplete fracture. Sambucus glycosides tablet were administered (each group at 220, 110 and 60 mg/kg doses) orally to adult Rabbits. After 12, 24, 36 days of treatment, animals were administration of marginal ear vein blood. Determination of the serum BGP, ALP, Ca 2+ and P changes with determination kit. By X-ray extent of fracture healing observed in each group. After the last dose, euthanized and femur bones collected, With DEXA bone mineral density tests were performed in order to determine bone mineral density (BMD), bone mineral content (BMC) and callus area. Detect TGF-β1, BMP-2, Col-Ⅰ a1 and Col-Ⅱ a1 mRNA expression in fracture healing each period using reverse transcription polymerase chain reaction and real-time fluorescence quantitative method. Results Sambucus glycosides tablet on fracture rabbit serum ALP, serum BGP, serum P levels were significantly increased, serum Ca 2+ concentration decreased; fracture zone of rabbit bone density, bone mineral content, bone callus area has significantly increased role. X-rays showed elderberry total glycosides tablet may promote healing of the fracture zone. Elderberry total glycosides tablet in fracture healing process of the BMP-2, TGF-β 1 had no significant effect, can promote the expression of Col-Ⅰ a1, inhibit the expression of Col-Ⅱa1. ConclusionSambucus glycosides tablet can promote rabbit fracture late maturation and calcification of bone matrix; promote collagen synthesis and deposition of calcium salts; improve quality of callus; accelerate fracture healing.

With bone fracture emerged as one of the most important health issues, more and more investigations are focusing on bone fracture healing. Fracture healing is a complex cascade of cellular events and the regulation of its healing remains poorly understood. Large number of clinical observations suggest that Sambucus treatment of fractures has a characteristics of short duration, rapid recovery and high efficacy.However, there are few studies on the mechanism of fracture healing.

Materials and methods

Fractures were made in the left tibiae of male New Zealand White rabbits weighing approximately 2.5 kg.The operations were performed under sterile conditions. The rabbits were anesthetized with Sodium pentobarbital. The fractures were made using the technique of Incomplete fracture.Sambucus glycosides tablet were administered (each group at 220, 110 and 60 mg/kg

doses) and traumatic pill (280 mg/kg doses)orally to adult Rabbits having a drill hole injury (2 mm) in the radial. Vehicle distilled water was given to the control group. After 12,24,36 days of treatment, animals were administration of marginal ear vein blood. Determination of the serum BGP, ALP, Ca2+ and P changes with determination kit. By X-ray extent of fracture healing observed in each group. After the last dose, euthanized and femur bones collected,With DEXA bone mineral density tests were performed in order to determine bone mineral density (BMD), bone mineral content (BMC) and callus area.Detect TGF-pi, BMP-2, Col- I ai and Col-Hai mRNA expression in fracture healing each period using reverse transcription - polymerase chain reaction and real-time fluorescence quantitative method[1-3].

Results and discussion

Compared with model group, Sambucus glycosides tablet high dose group, serum ALP 12 days in the treatment difference was significant (P <0.01); Sambucus glycosides tablet in the high dose group ALP medication 24 days at a high level, the difference was statistically significant (P <0.05).

Compared with the model group, Sambucus glycosides tablet high dose group, traumatic pill group administered 12 days in serum BGP were significantly differences (P <0.05); administered 24 days, Sambucus high total glycosides tablet , middle dose group, traumatic pill group has significant difference (P <0.01).

Compared with the model group, each group medication 12 days, serum Ca2 + no significant difference; medication 24 and 36 days, each Sambucus glycosides table group of Ca2 + levels with significant differences (P <0.05).

Compared with the model group, Sambucus glycosides table middle dose group medication 12 days, serum P at a high level, with a significant difference (P <0.01); medication 24 days , Sambucus glycosides tablet high dose, middle dose , low dose group and traumatic pill group P with a significant difference (P <0.05 or P <0.01); medication 36 days, Sambucus glycosides tablet high dose, middle dose and low dose group and traumatic pill group P a significant difference (P <0.05 or P <0.01).

Compared with the model group, Sambucus glycosides tablet each dose group fracture zone BMD, BMC, Area are at a high level, which Sambucus glycosides tablet each dose group was no difference in BMD; Sambucus glycosides tablet high dose, middle dose , low dose group and traumatic pill group there was a significant difference in BMC (P <0.01); Sambucus glycosides tablets dose group and traumatic pill group there was a significant difference between Area (P <0.05 or P <0.01). X-rays and microscopic observation showed Sambucus glycosides tablet may promote healing of the fracture zone.

Compared with model group, the treatment group TGFmRNA of expression were reduced. Compared with the control group, Col-1 a1mRNA expression in each treatment group were lower than the control group, with a significant difference (P <0.01); Compared with model group, 14 days, 21 days Col-1 a1 mRNA increased expression . Compared with the control group, Col-Ha1 mRNA expression in each treatment group were lower than the control group, 21 days treatment group with significant difference (P <0.01); Compared with model group, 14 days Col-Ha1mRNA expression volume increased.

Conclusion

Currently the ALP as a sign of osteoblast activity, ALP levels increased shown osteoblast activity expressed strong[4-6].BGP has a role in bone metabolism regulating hormones, BGP levels in the blood circulation and in bone content was positively correlated. Study found that calcium concentration is declining, phosphorus concentration increases, the peak was significantly increased calcium-phosphate product, promote calcium deposition, accelerate fracture healing [7]. Collagen is widely distributed in the extracellular matrix to form tissues and organs of the stent. Collagen as bone, cartilage, ligaments, tendons and other tissues of the basic structure of material, so that these organs have a high tensile strength [8].Sambucus glycosides tablet can promote rabbit fracture late

Abstracts:ObjectiveTo study the Sambucus total glycosides tablets analgesiaanti-inflammatory effects and its effects on fracture healing. Methodsanti-inflammatory effects by ear swelling in mice and rat foot swelling experiments; Analgesic action by mouse hot plate method; New Zealand white rabbit fracture modelthrough the light microscope and electron microscope observation of each degree of fracture healing.Results the total glycosides Sambucus piece of high, medium and low dose group to inhibit ear swelling in mice and rat foot swelling significantly inhibitory effect (P < 0.05).Sambucus total glycosides tablets of high, medium and low dose group in 1h can increase pain threshold of mice (P < 0.05). Sambucus total glycosides tablets of high dose group, medium and promote the role of bone tissue and osteoblasts to form. Conclusion Sambucustotal glycosides tablet has good resistance to acute inflammation and analgesic effect, and can promote the fracture healing.
