CHARACTERIZATION AND ANTIOXIDANT ACTIVITY OF MARINE MICROALGAE: AMPHORA SP. AND CHAETOCEROS SP. EXTRACTS

The characterization of lipid content and antioxidant capacity of the carotenoid astaxanthin from the marine microalgae, Amphora sp. and Chaetoceros sp. were investigated. In the present study, marine microalgae were surveyed and collected from the Eastern part of Thailand. Amphora sp. and Cheatoceros sp. were single growth and applied for screening of lipids content using the colorimetric sulfo-phospho-vanillin (SPV) method to monitor high lipid producers. The SPV method showed that Cheatoceros sp. had the higher lipid content of 0.63 ± 0.06 mg dry weight and followed by that of Amphora sp. of 0.37 ± 0.1 mg

dry weight. Moreover, the carotenoid as astaxanthin concentration in the microalgae extracts were analyzed using HPLC and the result presented that Chaetoceros sp. accumulates astaxanthin higher than Amphora sp. extract with 93.7 and 14.6 |ig/ml, respectively. However, the extracts of Amphora sp. exhibited strong antioxidant properties than Chaetoceros sp. with IC50 of 1,1 -dihpenyl-2-picrylhydrazyl (DPPH) radical at 0.05 and 0.34 mg mL-1, respectively. Our results suggested that marine microalgae might be a natural source of carotenoids as astaxanthin for human health and nutrition.

Extracts of the oleo-gum resin obtained from Boswellia serrata have been used since ancient times for the treatment of inflammatory diseases. The first purpose of this research was the characterization of seven B. serrata commercial extracts. The second aim was to investigate if B. serrata extracts can attenuate the deleterious effect of lipopolysaccharide (LPS) in an in vitro model. Six commercial dry extracts and a hydro-enzymatic extract were analysed by HPLC. Using analytical standards, it was possible to quantify 11-keto-p-boswellic acid (KBA), 3-O-acetyl-11-keto-p-boswellic acid (AKBA) and p-boswellic acid (pBA). The cytotoxicity of two different extracts (a dry extract and the hydro-enzymatic extract) and three analytical standards was evaluated using primary culture of porcine Aortic Endothelial Cells (pAEC) by MTT assay. HPLC analyses showed that different extracts from B. serrata contain different percentage of boswellic acids (KBA from 0.13 ± 0.01% to 4.61 ± 0.67%; AKBA from 0 to 3.83 ± 0.10%; pBA from 0.34 ± 0.02% to

11.56 ± 1.48%). Although all commercial extracts are titrated to 65% of boswellic acids, this percentage is unrealistic as evidenced by these results. The dry extract significantly reduced to 53.2 ± 2.7% cell viability, at the highest dose (100 ig/ml), while the hydro-enzymatic extract did not show cytotoxicity at any of the doses administered (from 2.4 to 2400 | g resin/ml). In response to LPS-induced stimulation, the dry extract significantly restored cellular viability only at lower doses (0.1 and 1 | g/ml), while the hydro-enzymatic extract was significantly effective at all doses studied. On the other hand, analytical standards, used at the same concentrations present in phytoextracts, did not decrease LPS-induced cytotoxicity. In conclusion, our results confirm the anti-inflammatory activity of B. serrata and emphasize the importance of the phytocomplex for the development of pharmacological action. However, attention should be paid to the composition of the commercial extracts.
