DHP1401 INHIBITS AMYLOID-BETA-INDUCED HIPPOCAMPAL LONG-TERM POTENTIATE DEFICIT VIA BDNF/TRKB SIGNALING

The seed of Zizyphus jujubavar.spinosa has been traditionally used for psychiatric disorders in Eastern Asia. Our recent findings have indicated thatthe ethanol extract of Zizyphus jujuba var. spinosa (DHP1401) improves memory impairment, a common symptom of various neurological diseases. However, the effects of DHP1401 on amyloid p (Ap) toxicity, which is a main cause of Alzheimer's disease, remain to be elucidated. In the present study, we examined the effect of standardized DHP1401 on Ap-induced synaptic long-term potentiation (LTP) deficit in the hippocampal tissue. DHP1401 blocked Ap-induced LTP deficits in a concentration-dependent manner. Moreover, DHP1401 increased brain-derived neurotrophic factor (BDNF)

level in naïve hippocampal slices. The finding that the blockade of BDNF receptor reduced the effect of DHP1401on LTP suggests that DHP1401ameliorates the Ap-induced LTP deficit through BDNF/tropomyosin receptor kinase B (TrkB) signaling. However, transcription or translation inhibitors failed to block the effect of DHP1401, suggesting that BDNF synthesis is not required for the action of DHP1401 on LTP. Finally, we found that DHP1401 stimulates plasmin activity. In contrast, plasmin inhibitor blocked the effect of DHP1401 on the Ap-induced LTP deficit. Our findings indicate that DHP1401 ameliorates Ap-induced LTP deficits through BDNF/TrkB signaling.

The aimof this study was to investigate the antioxidant and acetylcholinesterase (AChE) inhibitory activity of extracts from F. caspica M. Bieb. and F. halophile Pesmen by employing different in vitro assay systems. Antioxidant capacity of the obtained extracts were evaluated byABTS [1] and DPPH [2] radical scavenging activity, FRAP [3] and CUPRAC assays [4]. Total phenol contents of all extracts were also determined by using Folin-Ciocalteu assay and Anti-ChE activity was determined by Ellman method [5]. The results showed that the ethyl acetate extract from the aerial parts of F. caspica exhibited the highest phenolic content and antioxidant activity in all test assays with the value 177.23 mg gallic acid equivalent/g extract in Cuprac assay, 268.28 mg Trolox equivalent/g extract in ABTS and IC50 = 53.56 ^g/ml in DPPH radical scavenging assays and 207.38 mg quercetin equivalent/g extract in FRAP assay, respectively. On the other hand, the

methanol extract of F. caspica M. Bieb. the highest AchE inhibitory activity with the 0.04433 ^g/ml IC50 value when compared standard donepezil (IC50 = 0.00008 ^g/ml). However, AchE inhibitory activity of methanol extract which is higher than ethylacetate extract from F. caspica M. Bieb. could be due to a synergistic effect.
